The starting point of Line 6 is Xunfenggang in Jinshazhou, western Guangzhou. After crossing the North Ring Expressway, head southeast along the center of Jinshazhou Road. On the east side of Sha Feng Village, the Baisha River Bridge crosses the tributary of the Pearl River and connects with Shatou of Datansha Island. After that, the line turns due south, from elevated to underground tunnel, and then goes south to Shuangqiao Road to transfer to Line 5. After passing through Guangzhou-San Railway, the line obliquely passes through the tributary of the Pearl River, and Ruyifang Station is set at Duobao Road in the old Guangzhou South Railway Station. The line goes south along Huangsha Avenue to Huangsha Station of Datong Road and transfers to 1 line. After that, the line goes along the 623 road, passes through the Cultural Park and Renmin South Road, arrives at Haizhu Square along Yide Road, and then transfers to Line 2. After bypassing Guangzhou Jie Fangbei, pass Taikang Road, Wanfu Road and Yuexiu South Road in turn, pass a building near Donghua South Road and Dashatou Road, and arrive at Dongshan Lake Park. The tunnel passes through Dongshan Lake, turns northeast, along Dongshan Street, Guigang Avenue and Qianshu Road, and then transfers to Line 1. Then the line goes north along Nonglinxia Road, and changes again at Ouzhuang Station and Line 5. After that, the line turns into Xianlie Middle Road and Xianlie East Road with a small radius curve, then turns into Guangzhou Avenue North and Xinghua Road, and then transfers to the main line of Line 3 at Yantang Station. The line runs northeast along Yanling Road, changes to the branch line of Line 3 at Tianhe Bus Terminal, and finally reaches the end of Changba along Tianyuan Road.

Although Line 6 has not been completed and put into use, the planning and design of the branch line has been going on for a long time. The east extension of Line 6 (also known as the second phase of Line 6) from Changban Station along Tianyuan Road, Guangshan Road, Kaichuang Avenue to Luogang New City has been approved by the National Development and Reform Commission, and the overall design stage of the project is under way. The eastward extension will include Botanical Garden, Longdong, Kemulang, Gaotang, Huangpi, Jin Feng (formerly known as Xiangshan Road), Siam Gang (formerly known as Science City East), Suyuan (formerly known as Siam Gang), Luogang and Xiangxue *** 10 Station. In the planning, the eastern end will continue to extend eastward from Xiangxue and reach Yonghe via Xiangxue Park. At the same time, there are suggestions at the western end, extending from Xunfenggang Station to Feishui, Nanhai District, Foshan City.

Due to demolition and other reasons, Yidelu Station and Shahe Station were closed to traffic, and trains stopped passing through these two stations.
