(1) Disinfection of instruments. Glassware should be washed with washing powder before tissue culture. The inoculation room and clean workbench were scrubbed with 70% alcohol and irradiated with ultraviolet rays. Other utensils should also be sterilized at high temperature.

(2) Collection of culture materials. Plant materials used in tissue culture can be roots, stems, leaves, flowers, buds, cotyledons and hypocotyls of seeds, and sometimes pollen or anthers can be used. Usually, young materials should be taken, and these materials must be kept fresh when they are transferred to the culture medium, otherwise the tissue culture will fail.

(3) Disinfection of the culture. Rinse the collected materials with tap water first, then with distilled water for 2-3 times, then use sterile gauze to absorb the water on the materials, cut them into small pieces, soak them in 70% alcohol 15-30 seconds, then soak them in 30% bleach solution for 20 minutes for disinfection, and finally rinse them with sterile water for 3-5 times.

(4) preparing explants. Sterilize the sterilized materials with scissors and tweezers on the flame, peel off the scales of the bud, the outer skin of the bud and the endosperm of the seed coat on the sterilized filter paper, and then cut them into small pieces with a length of 0.2-0.5 cm (these small pieces are explants). During the operation, it is forbidden to touch these materials by hand.

(5) Inoculation and culture. Under aseptic conditions, the cut explants were immediately inoculated on the culture medium. Test tubes and triangular bottles used after inoculation should be sealed with sterile cotton wool and placed on the culture rack in the culture room for culture. Fluorescent lamps are used 12 ~ 16 hours every day. Keep it at 25℃ plus or minus 20℃. You can also use variable temperature culture, and the temperature at night is slightly lower than during the day.
