What is NSG mouse?

NSG mice are mice that directly knock out Rag2 and IL2rg genes in NOD/scid mice by using CRISPR/Cas9 system. Theoretically, there is no residual background, and the background is purer. Compared with NOD/scid mice, the survival rate of human cells and tissues transplanted in NSG mice is significantly improved, and a higher proportion of normal or cancerous human cells and tissues can be implanted.

In addition, NSG mice can meet the needs as a model of human immune system. After transplanting human hematopoietic stem cells, human T cells can be produced in the peripheral lymphoid tissue of NSG mice.

Research application:

It can be used as a transplantation model of human cells and organs and a tumor transplantation model, which is convenient for medical research, new drug development and toxicology experiments.

Pathogenic mechanism:

The IL-2RG gene encodes the IL-2 receptor γ protein, and some other IL-2 receptors cannot * * share this γ chain. However, the early development of T cells requires several IL-2 receptors, which will seriously affect the production of T lymphocytes.

The deletion of Rag2 gene hindered the development of lymphocytes. Therefore, when two genes are mutated at the same time, that is, Rag2-/- IL2rg-/-, B lymphocytes, T lymphocytes and NK cells in the body are missing, and the immune function is impaired.
